Última An in vivo test to assess mefloquine 25 mg/kg for the treatment of uncomplicated falciparum malaria in Rondônia, Brazil BJID Basano,S.A.; Bianco,A.; Taylor,W.R..J; Olliaro,P.; Camargo,L.M.A.. Drug-resistant Plasmodium falciparum is undermining malaria control efforts worldwide. In Brazil, mefloquine (MQ) at a dose of 15 mg/kg body weight is used to treat P. falciparum. At this dose, MQ resistance developed rapidly in Thailand. Use of a higher MQ dose may retard the development of resistance. We treated 50 patients aged one to 67 years who had acute, uncomplicated P falciparum malaria using MQ 25 mg/kg. There were no serious adverse events. Two patients complained of dizziness and insomnia. Assessing evaluable patients, the day 42 cure rate was 40/42 [95.2% (95% confidence interval 83.8 to 99.4%)]. Mefloquine was efficacious and well tolerated in this small cohort from the state of Rôndonia. The aim of this work was to study the effect of antimalarial drugs on the bioavailability of 99mTc-MDP in rats. Mefloquine (MQ) and artemisinin (AM) were administered in two treated groups (T) and sorbitol in control group (C) for 7 days. Then, 99mTc-MDP was injected in all groups and %ATI was calculated. A significant increase of %ATI in MQ group, from C to T, occurred in spleen (0.35±0.10to0.58±0.13), liver (1.69±0.28to3.31±0.07) and blood (0.79±0.17to2.09±0.53). The %ATI increased significantly in AM group:femur (2.76±0.59to5.98±0.70), liver (1.69±0.28to4.59±0.68), lungs (0.29±0.05to6.22±0.86), spleen (0.35±0.10 to0.86±0.15) and blood (0.79±0.17 to4.65 ±0.74). Abstract Objective To evaluate whether patient age has a significant impact on mefloquine concentrations in the plasma and erythrocytes over the course of treatment for uncomplicated falciparum malaria. Methods A total of 20 children aged between 8 and 11 years and 20 adult males aged between 22 and 41 years with uncomplicated falciparum malaria were enrolled in the study. Mefloquine was administered to patients in both age groups at a dose of 20 mg kg−1. The steady-state drug concentrations were measured by reversed-phase high performance liquid chromatography.
